CEInfinite couples directly to your preferred mass spectrometer for the rapid characterization of multiple product attributes. The addition of AES consumables enables outstanding reproducibility and sensitivity.

icIEF-MS enables rapid feedback on the post-translational modifications (PTMs) and other critical quality attributes (CQAs) underlying each protein charge isomer.

This can inform decision making during process development, and help mitigate unexpected setbacks during regulatory filing, scale-up, and manufacturing. Furthermore, it can help characterize ADCs, LNPs, another emerging modality that are challenging to characterize using other techniques.

With the introduction of icIEF-MS on the CEInfinite you can couple to a variety of mass spectrometers including the Orbitrap-based instruments for rapid intact mass analysis.

How Does icIEF-MS Coupling Work?

icIEF fractionation setup was converted into an icIEF-MS system by directly coupling the transfer capillary to a mass spectrometer. This enables intact mass analysis in 45 minutes or less for more rapid feedback on icIEF peaks.

By utilizing cartridges of 200 µm diameter, the resolution is prioritized, and importantly, acetonitrile and formic acid are added to the makeup solution to help prepare the icIEF peaks for electrospray ionization.

icIEF-MS Under Native Condition

Native mass spectrometry (nMS) is an emerging approach for studying large biomolecules and their complexes in the solution via the electrospray ionization mass spectrometry (ESI-MS), while maintaining as much as possible the native structural features of the analytes and their interactions in the gas phase during the electrospray. In biopharmaceutical development, native MS is a versatile method used to study complex protein heterogeneity with non-covalently driven assemblies in a native-like folded state.

AES develops a robust imaged capillary isoelectric focusing (icIEF) coupled to MS workflow under native condition, to characterize cysteine-linked antibody-drug-conjugate (ADC) charge heterogeneity. Successful separation and reliable identification of charge variants of latest-generation cysteine-linked ADCs including polatuzumab vedotin and brentuximab vedotin were achieved using icIEF-MS online platform under native condition. The developed icIEF-MS under native condition can be applied to the diverse complex proteins that are unstable under denaturing condition.
